摘要:
目的 探讨不同体重指数(BMI)分组的供体胰腺胰岛产量和功能差异。方法 纳入研究的93例胰腺均来自器官捐献者,根据亚洲人肥胖等级分组标准,将供体分为3组,分别为正常组(BMI:18.50~22.99 kg/m2)、超重组(BMI: 23.00~24.99 kg/m2)和肥胖组(BMI≥25.00 kg/m2)。利用Ricordi方法消化胰岛,连续密度梯度法纯化胰岛,检测胰岛分离效率、胰岛活性,葡萄糖刺激胰岛素分泌试验(GSIS)检测胰岛素分泌能力。结果 胰岛分离产量(IEQ)在肥胖组(BMI≥25.00 kg/m2)更高(F=3.05,P<0.05)。Pearson相关性分析显示供体BMI水平与胰岛功能呈正相关性(r=0.37,P=0.01)。结论 当供体BMI在一定范围内时,供体BMI水平越高,所得胰岛产量越高,且功能更好。
Abstract:
Objective To investigate the difference between the donors body mass index(BMI)and the islet isolation yield and function of islet.Methods The 93 cases of pancreas were collected for our study. According to the obesity classification standard in Asian, the donors were divided into three groups, including the normal group(BMI: 18.50-22.99 kg/m2), the overweight group(BMI: 23.00-24.99 kg/m2)and the obese group(BMI≥25.00 kg/m2). The method from Ricordi was used for digestion, and islets were purified by continuous density gradient method. The islets were identified by staining with the Dithizone(DTZ), the islets were analyzed for cell viability, and the insulin secretion ability of islets was determined by GSIS test.Results The islets yield(IEQ)in BMI≥25.00 kg/m2 group was more than other two groups(F=3.05, P<0.05). Pearson analysis showed that the level of donor BMI was positively correlated with islet function(r=0.37, P=0.01).Conclusion The higher level of donor BMI may be associated with more islets yield and better islet function in certain range of isolation BMI.
