摘要:
脱碘酶是含硒蛋白酶,包括一型脱碘酶(D1)、二型脱碘酶(D2)和三型脱碘酶(D3),它们在甲状腺激素代谢过程中起重要调节作用。其中D2主要在T4向T3转化、rT3向T2转化过程中起重要作用。D2泛素化过程中涉及两种泛素化连接酶:WSB1、TEB4; 两种去泛素化酶:USP20、USP33。D2对下丘脑-垂体-甲状腺轴调节甲状腺激素的产生过程起重要作用,在甲状腺功能减退症及其治疗中有重要影响,其表达还与甲状腺髓样癌有关。D2泛素化与慢性碘过量致促甲状腺激素水平升高有关。同时,D2基因多态性参与甲状腺激素代谢,影响甲状腺激素水平。
Abstract:
Deiodinase is a selenoproteinase, including type 1 deiodinase(D1), type 2 deiodinase(D2)and type 3 deiodinase(D3), which plays an important regulatory role in the metabolism of thyroid hormone. D2 plays an important role in the transformation from T4 to T3, from rT3 to T2. There are two types of ubiquitination ligases such as WSB1 and TEB4, and two deubiquitination enzymes such as USP20 and USP33, involved in the process of D2 ubiquitination. D2 plays an important role in the regulation of thyroid hormone by hypothalamic-pituitary-thyroid axis(HPT), and plays an important role in hypothyroidism and its treatment. Its expression is also related to medullary thyroid cancer. D2 ubiquitination also involves in the elevation of thyroid stimulating hormone induced by chronic iodine overdose. Meanwhile, D2 gene polymorphism is involved in the metabolism of thyroid hormone and affects the level of thyroid hormone.
